Heart attacks: Naked mole-rats bear lifesaving clues
ScienceDaily (Feb. 23, 2012) ? Could blind, buck-toothed, finger-sized naked mole-rats harbor in their brain cells a survival secret that might lead to better heart attack or stroke treatments?
University of Illinois at Chicago biologist Thomas Park and colleagues at UIC and the University of Texas Heath Science Center at San Antonio think the subterranean lifestyle of the pasty-looking rodents may indeed hold clues to keeping brain cells alive and functioning when oxygen is scarce. The key may lie in how brain cells regulate their intake of calcium.
"Normally, calcium in brain cells does wonderful things, including forming memories," says Park, who is professor of biological sciences at UIC. "But too much calcium makes things go haywire."
Brain cells starved of oxygen can't regulate calcium entry, and too much calcium in the cell is lethal. When a heart attack or stroke prevents oxygenated blood from reaching the brain, brain damage or death results.
Naked mole-rats, however, are very tolerant to oxygen deprivation, or hypoxia -- as are human newborns, whose brain cells have calcium channels that close during oxygen deprivation, protecting the cells from calcium overdose. With age, these calcium channels no longer close, which normally isn't a problem -- except during a heart attack.
Naked mole-rats retain a tolerance for oxygen deprivation into adulthood. Park and his colleagues measured calcium entry in brain tissue that had been kept under oxygen-poor conditions, reporting their findings online Feb. 21 in PLoS One.
"We knew the adults of this unusual mammal had brains that, like infant humans, were very tolerant to oxygen deprivation," he said. "We wanted to know if the adult naked mole-rats used the same strategy as babies to prevent calcium entry. This is exactly what we found."
Park thinks this strategy is an evolutionary adaptation by mole-rats, which live in the hundreds underground in tight, oxygen-deprived conditions.
"Imagine 200 mice living in a shoe box buried four feet under the ground -- things are going to get bad fast," he said.
The researchers think they have identified a novel mechanism for protecting the adult brain in times of oxygen deprivation.
"Developing this target into a clinical application is our next goal," he said. "We need to find a way to rapidly up-regulate the infant-type of calcium channels. Adult humans actually have some of these channels already, but far fewer than infants."
Park, who for years has studied naked mole-rats and their unusual adaptations, thinks the latest findings "are just the tip of the iceberg" of what we can learn from the rodents. Their homes are not only oxygen-poor, but rich in carbon dioxide and ammonia -- conditions that would make most animals ill. Yet mole-rats have evolved to suppress pain and even cancer.
"The more we study these creatures," said Park, "the more we learn."
Co-authors include Rochelle Buffenstein, of the University of Texas Health Science Center in San Antonio; Bethany Peterson, a UIC doctoral student in Park's lab; John Larson, UIC associate professor of psychiatry; and Christopher Fall, UIC visiting research associate professor of bioengineering.
The study was funded by the National Science Foundation and the National Institutes of Health-National Institute of Mental Health Neurotechnology Program.

COLUMBUS, Ohio (AP) ? John Glenn made his historic spaceflight alone in 1962 but celebrated its 50th anniversary Monday among hundreds of people within his orbit, from fellow headline-making astronauts and NASA's administrator to family, friends and students at Ohio State University, where the public affairs school bears his name.
They watched footage of the launch, laughed at his enduring sense of humor and showered him with applause, praising the man who became the first American to orbit the Earth on Feb. 20, 1962.
Neil Armstrong, the first man to set foot on the moon, told the audience at the celebratory gala that Glenn was "no ordinary pilot." There was a need for leadership in the space program in the early 1960s, Armstrong said, and Glenn "literally rose to the occasion."
The former astronaut and U.S. senator from Ohio, now 90, circled the Earth three times in five hours and was viewed as a national hero for helping to lead the United States into space.
"I think the hero thing is in the eye of the beholder," Glenn said during a question-and-answer session with Annie, his wife of nearly seven decades. "I don't look at myself that way."
Asked about his heroes, Glenn said he admires different qualities in different people, such as the perseverance of former U.S. Rep. Gabrielle Giffords of Arizona, who was severely injured in a shooting last year.
Her husband Mark Kelly, a retired astronaut and commander of the space shuttle Endeavour's final mission, was the night's featured speaker and said he was honored to be sitting between two of his own heroes, Armstrong and Glenn. He brought the audience a message from Giffords: "Be passionate. Be courageous. Be strong. Be your best."
Glenn urged the audience to support research and education and shared the lessons he learned when he was among the top military test pilots presented in 1959 as the Mercury Seven. The only other surviving Mercury astronaut is Scott Carpenter, who called out the memorable line "Godspeed John Glenn" moments before the rocket ignited for Glenn's spaceflight.
"With a lot of work and a lot of people organized, you can do almost anything," Glenn said.
Earlier Monday, NASA had surprised Glenn with the kind of anniversary gift only a space agency can give, enabling him to speak live with the International Space Station from a stage at Ohio State University.
Sitting on stage with NASA Administrator Charles Bolden, he chatted with three space station crew members about space research and NASA's future. Commander Dan Burbank appeared by video link, flanked by two flight engineers floating in the zero-gravity environment, and said the crew was delighted to help commemorate Glenn's momentous trip.
"Fifty years ago today, Friendship 7 was orbiting planet Earth, and that helped in a very big way, paved the way for America to become a space power, and to go to the moon, and to do the things that we're doing right now on the International Space Station," Burbank said.
Glenn had a light-hearted but educational exchange with the crew, asking them about the types and number of experiments on board ? more than 100, they said ? and explaining to his gravity-bound audience of more than 200 people that, for example, a candle burns differently in space than on Earth.
When Bolden asked the astronauts which experiment they'd like to hand off to Glenn if he could join them, Burbank suggested research on the "regenerative environmental control systems" on spacecraft.
"That's a fancy word for our toilet," flight engineer Don Pettit added. "So he wants to put Sen. Glenn busy fixing the plumbing up here."
Glenn took the humor in stride, replying: "That's exactly what I thought I was going to get assigned to."
Bolden joked that Glenn sometimes bugs him about making a trip to the space station. Glenn became the oldest person to fly in space in 1998, at age 77.

Billionaire Falcone: no LightSquared bankruptcy (Reuters)
(Reuters) ? Hedge fund manager Philip Falcone is ruling out a bankruptcy filing for his telecom startup LightSquared Inc, one day after U.S. regulators said they planned to revoke approval for the company to build a national wireless broadband network.
"It is clearly not on our table," Falcone said in an email to Reuters on Wednesday when asked if LightSquared, the biggest investment in his Harbinger Capital Partners fund, was considering a bankruptcy filing.
He said there is a plan for dealing with the Federal Communications Commission's plan to rescind its permission for LightSquared to build out a land-based network, but he declined to offer any details.
LightSquared said the FCC's action had harmed it and the public by preventing construction of a network vital to U.S. competitiveness.
Industry analysts and some LightSquared investors say the telecom startup is running short of options with cash draining away and little chance of getting the FCC to change its mind. They also say it could be hard to find a buyer for LightSquared. Several suggested that a bankruptcy filing, while perhaps not imminent, is the likely outcome.
In a financial filing last year, LightSquared said it could run out of money by the middle of this year.
"A bankruptcy Chapter 11 filing seems inevitable," said a person with one of the hedge funds that owns some of LightSquared's more than $1 billion in outstanding debt. The person declined to be named because the situation is still fluid with LightSquared.
Tim Farrar, a veteran industry analyst and principal at TMF Associates, agreed but said a bankruptcy filing may not occur immediately. "Investors don't have leverage to force a filing right now," he said, adding that a final FCC ruling would strengthen the hand of investors to force it.
The FCC announced its decision late on Tuesday, minutes after the National Telecommunications and Information Administration said LightSquared's network would interfere with the Global Positioning System, vital to the military and with civilian uses ranging from airline navigation to irrigation.
Many telecommunication analysts have said it will be difficult for the financially-strapped company to either raise additional financing or ever become profitable with just the satellite-based portion of its network.
Its plan had been to build a high-speed wireless service so it could tap into the massive demand for data on devices like smartphones and tablet computers.
"We remain committed to finding a solution and believe that if all the parties have that same level of commitment, a solution can be found," LightSquared Chief Executive Sanjiv Ahuja said in a statement without tipping the company's next move.
HARBINGER SUFFERS
The fate of LightSquared also is critical investors in Falcone's hedge fund, which once controlled $26 billion in assets but is now down to about $4 billion. A little more than half of Harbinger's money is tied-up in LightSquared and the hedge fund is the company's single largest equity investor.
Last year, Harbinger, a fund known for its volatile returns with years of big losses often following year of big gains, gave up 47 percent of its value in large part because of a sharp markdown on the value of the LightSquared investment.
Falcone, an expert in distressed investing whose well-timed bet against the overheated housing market earned him a big following in the $2 trillion hedge fund industry, still has a prominent roster of investors. The list is said to include hedge fund legend George Soros, who declined to comment on the Falcone's current troubles.
For now, LightSquared's main business partner Sprint Nextel is sticking to its plan to give the company until the middle of next month to try and come up with a solution.
"We have an agreement with LightSquared that goes to March 15 to give LightSquared time to address the issues it's had with the FCC," Sprint spokesman Bill White said.
Sprint and LightSquared already decided at the end of last year to try and save money by shelving work on the network until the situation was clearer.
If Sprint scuttles the deal it could mean money coming back to LightSquared. "There is $65 million that would be returned to LightSquared to reflect work that was paid for (but) not carried out." White said.
LightSquared has argued that its network would help relieve a national wireless capacity shortage, in line with FCC goals to devote more airwaves to mobile services.
But the GPS interference and the FCC's subsequent decision may pose too great an obstacle to overcome, analysts said.
"While we could not commit to saying that it is the 'end of the road' for LightSquared, we believe it will be an extremely tough slog for the company to show value in the spectrum it holds," Wells Fargo analyst Jennifer Fritzsche wrote in a research note.
OPTIONS AND ALTERNATIVES
One route would be for LightSquared to consider suing the government if the FCC's final determination goes against the company after the comment period ends March 1.
"The odds are against (LightSquared) in being able to persuade the FCC that it is wrong," said Andrew Lipman, a partner who heads law firm Bingham McCutchen's telecommunications practice.
LightSquared would likely have to wait until the comment period is over before it could sue the FCC and such a case would likely take about nine months or more, the lawyer estimated.
Another way to deal with the GPS issues would be to arrange a spectrum swap with another carrier or company, a person close to LightSquared who is not authorized to speak publicly said.
A sale of LightSquared might also be possibility, one investor in Falcone's fund and other industry analysts have suggested, but they acknowledge that it might be tough to find a buyer.
"Right now (LightSquared is) in a world of hurt and the thing is it is toxic enough that no one wants to take it," said one telecommunications mergers and acquisitions expert who could not speak about the matter publicly.
This person added that companies like AT&T and Verizon have been interested in snapping up spectrum, but said buying LightSquared would not be a desirable way to do so, considering the complications around it.
The value of LightSquared's spectrum declines significantly if it cannot be used for cellular services, analysts have said.
For Falcone, the regulatory set-back is the latest in a string of problems dogging the 49-year old former Harvard hockey star who grew up in rural Minnesota but now lives on Manhattan's
Upper East Side in a townhouse once owned by Penthouse publisher Bob Guccione.
The U.S. Securities and Exchange Commission recently warned Falcone and some of his key lieutentants they face questions over whether they might have engaged in improper trading. Falcone has also faced investor anger over having failed to tell them that he took a loan from the fund to pay his personal taxes at a time they could not get their money out.

Dan Chaon's 6 favorite collections of terrifying tales (The Week)
New York ? The award-winning author admires spooky stories by Edith Wharton, Ray Bradbury, and Joyce Carol Oates
The Ghost Stories of Edith Wharton (Scribner, $17). Having been forced to choke down Ethan Frome in high school, I resented Wharton until I discovered her mysterious and coolly menacing stories of the supernatural. My favorite is the beautifully subtle shocker "Afterward," in which Mary Boyne is told that her house is haunted ? but that she won't realize she's seen a ghost "till long, long afterward."
The October Country by Ray Bradbury (Del Rey, $8). Reading these stories is like visiting a Midwestern county fair after midnight, knowing that something bad is lurking on the darkened midway. Though Bradbury is thought of as a science fiction writer, I think these dark fantasy tales are his real achievement.

Blow-Up and Other Stories by Julio Cort?zar (Pantheon, $15). I love the way that the uncanny slowly encroaches into the world of Cort?zar's characters. The story "Blow-Up" was the basis for Michelangelo Antonioni's mod, groovy Hitchcockian film of the same name. Other stories, like "Axolotl" and "Continuity of Parks," are even more deeply hallucinatory and unsettling.
Will You Please Be Quiet, Please? by Raymond Carver (Vintage, $15). Just because the ghosts don't actually materialize out of the shadows doesn't mean they aren't there. Carver is classified as a realist, but the stories in his first collection are infused with such a strange, ambient sense of dread that they seem like ghost stories by default.

Haunted: Tales of the Grotesque by Joyce Carol Oates (out of print). Oates can do anything, but this collection of unsettling psychological horror stories distills some of her best tricks. "Extenuating Circumstances" burrows so deeply into the character of a terrifyingly crazy mother that you want to hide your eyes at the end.
Open Secrets by Alice Munro (Vintage, $15). Munro is a naturalistic writer if there ever was one, but she's fascinated by mystery, too. As the title suggests, these stories explore the unknown and unexplainable in the lives of ordinary people, and show how truly mystifying and spooky the workings of the human heart can be. There's at least one (maybe two) actual ghosts tucked into these pages, too.

Regarding this body and soul relationship conversely, the sexual act is designed only to express the spiritual reality of marriage.? Sex communicates to the spouse a love of exclusivity, permanence, completeness and fruitfulness.? Each time spouses engage in sexual intimacy, the vows of marriage are restated through this physical expression.? Naturally, as a bride or groom would not fail to make one of these promises on their wedding day, the promises must be held intact throughout the marriage.? For this reason, as stated above, the use of contraception causes the body to communicate to the spouse falsely; that a fruitful love exists in the sexual expression of the marital vows when it does not.
